Given these are the Guardians of the Twelveswood and aren't diseased in any way, I do wonder why Gladiators have to defeat some Treants. Blame the Calamity for altering their behaviour - that does seem to be the easiest excuse!
Treant Location
These are found in throughout the Snakemolt "pass" between Quarrymill and Rootslake. If you haven't unlocked Amdapor Keep yet, then you can hit two Treants with one stone. Guess you'd still need to defeat the Treants, now that you've annoyed them with stones. Use your Gladiator skills instead then?
Aetherytes
While I think, technically Quarrymill is the closest, if you're already near Camp Tranquil then don't bother teleporting to Quarrymill.
